ABSRACT

Single Cell Mass Cytometry (CyTOF) is a next generation flow cytometry platform which utilizes elemental mass spectrometry to detect metal-conjugated antibodies that are bound intracellularly or extracellularly to antigens of interest on single cells. Using elemental mass tags to replace fluorescent mass tags, CyTOF realizes over 50 protein co-analysis on single cells. With integration of micro-fluidic systems, CyTOF technique permits systematic evaluations of multi-complex biomarkers with trace amount of clinical samples. Our lab is among the first few groups established CyTOF technique in China. Meanwhile, changing the course of how diseases are treated and ultimately cured requires a comprehensive understanding of complex cellular phenotypes and their interrelationships in the spatial context of the tissue microenvironment. Imaging Mass Cytometry (IMC) using metal-tagged antibodies empowers simultaneous imaging of up to 37 protein markers at a time. Bringing together high-parameter CyTOF technology with imaging capability, the IMC technique enables deep interrogation of tissues and tumors at subcellular resolution to uncover pathology insights, new biomarker correlations and cell interactions.

单细胞质谱流式技术(CyTOF)是一种新一代的流式细胞分析技术。它利用元素质谱技术检测细胞内或细胞外与单个细胞相关抗原结合的金属耦合抗体。CyTOF用元素质量标签代替荧光质量标签,实现了单个细胞50多个蛋白质的共分析。随着微流控系统的集成,CyTOF技术允许对多个复杂的生物标志物进行系统的评估。我们的实验室是国内首批建立这项技术的课题组之一。同时,改变疾病治疗和最终治愈的过程需要全面了解复杂的细胞表型及其在组织微环境的空间环境中的相互关系。使用金属标记抗体的成像质谱流式细胞分析仪(IMC)可以同时在一个病理切片上成像多达37个蛋白质标记物。IMC技术将高参数细胞因子技术与成像能力结合在一起,能够在亚细胞分辨率下对组织和肿瘤进行深入的检测,从而揭示病理学规律、新的生物标记物相关性和细胞间相互作用。

BIOGRAPHY

Dr. Xianting Ding is Professor of Shanghai Jiao Tong University (SJTU), Department Head of Molecular and Nanomedicine Department for School of Biomedical Engineering, Deputy Director of Institute for Personalized Medicine. He was elected into the Chinese Top Elite Researcher Program, Qiushi Foundation Elite Young Scientists, World Economic Forum (WEF) Young Scientists, and World Laureates Forum (WLF) Young Scientists. He recently received Microsystems & Nanoengineering 2020 (MINE2020), Young Scientists Award. His research interests focus on developing Biomedical Micro-systems for single cell protein analysis with trace clinical specimens for precision and personalized medicine.
 
Dr. Ding has devoted in Biomedical Micro-systems for single cell based protein analysis and clinical translational applications. His lab is amongst the first few groups in China to establish single cell mass cytometry (CyTOF) technique and imaging mass cytometry (IMC) technique. His lab is also the pioneer lab in China to establish single cell western blot (scWB) technique. The lower detection limit of protein detection reported from his laboratory is amongst the most sensitive technologies based on PubMed and Web-of-Science retrieval. These cutting-edge technologies have been successfully applied for clinical disease diagnostics and therapeutics to benefit thousands of patients.

丁显廷,上海交通大学教授,生物医学工程学院分子与纳米医学系主任,个性化医学研究院常务副院长。先后入选“国家特聘专家专家”青年项目、求是基金会“求是杰出青年奖”、世界经济论坛(World Economic Forum, WEF)“青年科学家”、世界顶尖科学家论坛(World Laureates Forum, WLF)"青年科学家"、上海市科协“飞翔计划”。他最近获得了Microsystems & Nanoengineering 2020 (MINE2020), Young Scientists Award。他的研究兴趣集中在开发生物医学装置,用于单细胞蛋白质分析和微量临床样本的精确和个性化治疗。

丁博士致力于生物医学系统的单细胞蛋白质分析和临床转化研究。他的实验室是中国最早建立单细胞质谱流式技术(CyTOF)和成像质谱流式技术(IMC)的课题组之一。他的实验室也是中国建立单细胞免疫印迹(scWB)技术的先驱实验室。他的实验室对于临床痕量样本蛋白质检测精度是国际上目前有报道的技术中最灵敏的技术之一。这些前沿技术已成功应用于临床疾病诊断和治疗,使成千上万的患者受益。
